Initiation And Characterization Of A New Cell Line From Spodoptera Littoralis (New Sl)

Abstract


A new cell line was established and cloned from pupal ovaries of Egyptian cotton leaf worm Spodoptera littoralis and characterized at the morphological level, growth, and karyotyping level. The chromosomal number was compared to a reference cell line Sf9 and to eggs of the same insect. Cells were spherical at the beginning but some of them turned to be spindle shaped, maximum cell population found to be 4.8x106 at the logarithmic phase, cell doubling time was 24hr. Chromosomes were typical to those described from other species of Lepidoptera. Chromosomes number was counted on 57 metaphase for the new cell line (new Sl) and it ranged between 4-203 Chromosomes where as 23% of counted metaphases contained 6 chromosomes only. As for 58 metaphases were prepared from eggs, the counted chromosomal number ranged between 4-24 on 57 metaphases and only one metaphase had 46 chromosomes, but 20% of metaphases showed 8 chromosomes. The obtained results for eggs and new Sl was different from Sf9 which showed chromosomal number range from 61- 263 in 68 metaphase,; from which 30% was ranged from 103-120.
