Virogenesis In New Established Spodoptera Littoralis Cell Line(Sl 96), Adapted At 19 C Permissive For Spli.Granulovirus, Phop.Granulovirus, And Spli.Nucleopolyhedrovirus

Abstract
A complete replication of the Spodoptera littoralis granulovirus (SpliGV), nucleopolyherovirus (SpliNPV), and Phthoremea operculella granulovirus (PhopGV) was obtained in vitroby both viral infection and DNA transfection in the S.littoralis (Sl96) cell line which was established and completely characterized from ovariouls tissues of the cotton leaf worm, Spodoptera littoralis. SpliGV and PhopGV multiplied significantly during several passages in Sl96 cells at 19°C. Cells were infected and kept at 19°C for the first 4 hours post -infection. Then all the infected cells were incubated at 27°C till end of the experiment (20 days). The viral multiplication proceeded at the same rate. Comparison of SpliGV progenies multiplied either in vivo or in vitro, using electron microscopy and restriction profile analysis, showed their identity. Viral infections were detected also by specific nucleic probes. |
