Production Of Insect Resistant Potato By ýAgrobacterium Mediated Transformation With Cry1ac ýGene From Bacillus Thuringiensisýý

Abstract


In Oman, potato cultivation has increased in recent years as consumer ýdemand for potato products has risen. Tuber moth (Phthorimaea ýoperculella) is a serious constraint to increase the potato production in Oman. ýThe insecticidal proteins, (-endotoxins) are specifically lethal to Lepidopteran ýinsects. An attempt is being made to transform a potato cultivar with Bacillus ýthuringiensis gene which codes for an insecticidal protein. In order to express ýendotoxin in transgenic potato, a cry1Ac coding sequences from B. ýthuringiensis has been constructed into Agrobacterium vector (pRD400) with ýdouble 35S AMV promoter suitable for dicot transformation using kanamycin ýresistance gene as marker. Transgenic callus of potato has been grown on modified ýMS medium containing 300 mg/L carbenicillin and 150 mg/L kanamycin. The ýintegration of cry1Ac gene into potato genome has been verified by PCR ýamplification.ý
